If you upload some files and create a new item, it should generate derivatives for all of the files there at the creation. However if you then add additional files to the item it does not seem to trigger a derive for those. However you can do this yourself by clicking Edit Item at the bottom left of the item page, choosing "change the information", clicking the "Item Manager" link at the top of the page, and then clicking the "derive" button. (I think this is available to non-admins.)
